monoclonal antibodiesMonoclonal antibodies are essential tools in the work of Cancer Research UK, for both research and clinical applications. The Production of monoclonal antibodies is time consuming and very specialised. The Monoclonal Antibody Service (MAS) is a central resource available to all Cancer Research UK labs and grantees, providing technlogical expertise tailored to the immunological needs of our scientists.

Monoclonal Antibody Production

We have been supplying the London Research Institute and Cancer Research Technology for many years and have built up an extensive database of antibodies. We can also purify polyclonal antibody from serum supplied from immunized animals, and subclass your hybridomas by ELISA.

Antibodies are usually purified using Affinity Chromatography. The sample is eluted using a commercially bought Neutral Elution Buffer and exchanged into PBS using a G25 column. The concentration of the purified antibody will vary from sample to sample, usually between 0.8 and 1.2 mg/ml.

All growing Hybridoma cultures are tested for mycoplasma infection, and ELISA tested to determine subclass and activity. Purified monoclonals are reduced and screened on a gel to determine the integrity of the antibody. The results of these tests can be sent to you if required.

For purification of antibodies by affinity chromatography, we can supply protein A and G sepharose at a fraction of the cost of buying direct from commercial suppliers.

Hybridoma Fusion

The Hybridoma Fusion section is able to make novel antibodies from specific antigen. These antibodies can be used for basic research in a variety of assays, for co-localisation studies or in identification of their biological activity in vivo. Novel antibodies may be used in clinical or commercial applications. All our antibodies are ELISA positive and tailored to the particular application needed.
