Cytometers

We currently have eight cytometers located at 44LIF in Rooms 110-112 and a ninth (the LSC) is situated in Room 121. All analytical cytometers may be booked via the web-based booking system. You will be able to view sorter appointments but all bookings MUST be made via one of the staff of the FACS Lab.

Calibur 1

FACS Calibur 1 (Becton Dickinson)

This is used for analysis only.  It is a dual laser system.  The primary laser is an air-cooled 15mW argon ion laser emitting at 488nm, thus allowing two light scatter parameters and three fluorescence channels to be measured.  The second laser is a red diode laser emitting 635nm thus allowing the excitation of other dyes such as allophycocyanin or TO-PRO-3.

FACS Calibur 2 (Beckton Dickinson)

LSRII (Becton Dickinson)

This is used for analysis only and is equipped with four lasers.  These are UV (355nm), violet (407nm), blue (488nm) and red (635nm). The configuration is six colours from the blue laser, three from the red laser and two each from the violet and UV.

LSRII (Becton Dickinson)

LSRII (Becton Dickinson)

This is used for analysis only and is equipped with four lasers.  These are violet (407nm), blue (488nm), yellow (561nm) and red (635nm). The configuration is three colours from the blue laser, three from the red laser and six each from the violet and yellow lasers. This cytometer is very useful for excitation of QDot-labeled antibodies and red variant fluorescent proteins.

Laser Scanning Cytometer (CompuCyte)

Laser Scanning Cytometer (CompuCyte)

The laser scanning cytometer is a slide-based cytometer that gives similar information to our flow cytometers.  It is equipped with an air-cooled argon ion laser emitting at 488nm and an air-cooled Helium-Neon laser emitting at 633nm, allowing the simultaneous measurement of up to four colours or three colours plus light scatter.

FACSArray

FACSArray (Becton Dickinson)

The FACS Array is designed to be a bead analyser. CBA (Cytometric Bead Array) technology allows 'cell-free cytometry': capture beads are used to bind secreted molecules such as cytokines and by visualising these with a fluorochrome-labeled antibody picogram quantities of analyte can be measured. It uses a 96-well format and is also suitable for measuring fluorescence from cells.

The FACSArray is user-operated (after instruction) and is available for use 24 hours per day (subject to approval!).

MoFlo (BeckmanCoulter)

MoFlo (DakoCytomation, now BeckmanCoulter)

The MoFlo is a high pressure, 4-way cell sorter. This cytometer has a three laser set up.  There are three water-cooled Innova lasers. Two Innova 70c argon ion lasers tuned to 488nm and multiline UV and an Innova 90 Krypton laser capable of generating 568nm and 648nm lines. Up to 9 fluorescence parameters may be measured. Use of the sorters is by trained operators only!

MoFlo (BeckmanCoulter)

MoFlo (DakoCytomation, now BeckmanCoulter)

The newer MoFlo also has a three laser set up - a Coherent solid state 200mW 488nmlaser, a 633nm laser diode and a 200mW JDSU XCyte UV laser. It is capable of measuring up to nine fluorescence parameters and is also fitted with CyClone, the automated deposition unit.

FACS Aria (Beckton Dickinson)

FACS Aria (Becton Dickinson)

The Aria is also a high speed, 4-way sorter.  It has three solid state lasers emitting at 407nm, 488nm and 633nm.  It is a digital system capable of high speed 4 way sorting, single cell sorting and analysis/sorting of up to 12 fluorochromes.

In addition, we have several computers (Macs) that can be used for offline analysis.  Programs available include FlowJo, ModFit and FCS Express.
